3D Visualization of Retinal Vascular Pericytes in Mice by Immunostaining

전체 마운트 망막의 외부에서 볼 때, 조직 제거 처리를 받은 경우 조직 제거 처리가 없는 망막에 비해 전체 마운트 망막의 조직 투명도가 증가했습니다(그림 1).
산 망막 전체에 대한 조직학적 검사를 위해 후안와 주사를 통해 망막 혈관 구조를 형광 토마토 렉틴으로 빨간색으로 표시하고, 망막 주위 세포는 PDGFR-β로 녹색으로 표시했습니다(그림 2, 그림 3 및 그림 4). 이러한 라벨링은 조직 제거 전후에 샘플에서 관찰할 수 있었지만, 이에 비해 조직 제거 처리로 인해 망막 샘플의 배경 소음이 감소했습니다(그림 2). 이 치료법은 또한 파노라마 조직 절편 스캐너(그림 2)와 레이저 스캐닝 컨포칼 현미경(그림 3)에서 표지된 망막 주위 세포와 혈관 구조를 더 선명하게 만들었습니다.
망막 주위 세포의 점진적 분포는 또한 망막 혈관 나무를 따라 입증되었습니다(그림 3). 망막의 중앙 부분에서 말초 영역까지, 주피세포는 혈관 몸통과 분지, 모세혈관을 포함한 망막 혈관 구조의 기저막에 위치합니다(그림 3).
위의 관찰을 바탕으로 주피세포와 망막 혈관 구조의 공간적 상관관계는 투명화된 망막 샘플의 더 높게 확대된 Z 스택 이미지로 추가로 평가되었습니다(그림 4). 이미지 처리 시스템의 도움으로 3D 뷰를 추가로 제공하여 주피세포가 서로 다른 직경의 망막 혈관 구조를 단단히 감싸고 있음을 보여주었습니다(그림 4). 그림 4A0-D0 은 망막 혈관이 가장 높은 곳에서 가장 낮은 곳으로 계층적으로 분기되는 것을 나타냅니다. 주피세포는 모세혈관에 더 풍부하게 분포되어 있기 때문에 그림 4D0 의 PDGFRβ 염색은 그림 4A0-C0에 비해 더 강한 강도와 신호를 보여줍니다.
조직 제거 처리가 없는 마우스 망막과 대조적으로(그림 1A, 그림 2A, 그림 3A0-D0), 현재의 접근 방식은 전체 마운트 망막에서 주피세포와 혈관 사이의 공간적 관계를 보여주기 위한 망막 배경 신호를 효과적으로 줄였습니다(그림 1B, 그림 2B, 그림 3A1-D1).
